概述

   甘草作为一种常用中药,已被人们接受和使用,其主要成分是:甘草甜素、甘草甙、甘草类黄酮、后幕比檀素、刺芒柄花素、槲皮素等。具有解毒,抗炎,镇咳,抗肿瘤,抗溃疡,抗菌等作用。同时,甘草甜素对艾滋病病毒具有抑制增殖作用;甘草次酸对骨髓瘤及腹水肝癌均有抑制作用。甘草酸具有明显的抗利尿作用;甘草素、异甘草素具有抗溃疡和解痉作用;甘草黄酮类尚具有抗氧和抑菌作用。国家药典认定的甘草药材原植物有三种,即甘草(Glycyrrhiza uralensis Fisch. )、胀果甘草(Glycyrrhiza inflata Bat. )和光果甘草 (Glycyrrhiza glabra L.) 的干燥根及根茎。其中以甘草分布最广,产量最多,质量最好。甘草是我国传统中药材,应用历史悠久,名扬海内外。早在战国时期,就有利用甘草治病的记载,距今己有2500多年。东汉《神农本草》称甘草为“美草”和“密甘”,列为上品。中医药认为:甘草味甘、平,是补脾益气,止咳祉疾,缓急止痛,调和诸药,解毒的良药。美国FDA将甘草提取物列为安全无毒物质。科学研究证明,甘草主要含有甘草酸和黄酮类甘草甙(甘草甜素)。甘草提取物是一种具有抑菌、消炎、解毒、除臭等多种功能的添加剂

甘草酸物理化学性质

甘草末为淡黄色粉末,味甜,略带苦味,含6%~14%的甘草酸。纯品甘草酸为结晶性粉末,味极甜,甜摩为蔗糖的200~250倍,甜味存留时间长。熔点212~217℃。易溶于热水,不溶于醚,难溶于丙二醇、乙醇。对热、碱和盐稳定,pH 54、于3时,溶液会出现沉淀。小白鼠经口LD50大于10g/kg。ADI延期确定(FAO/WHO,1994)。

甘草酸产品用途

用途一:甘草是我国传统的调味料和中药,安全性高。有特殊的风味,与蔗糖、糖精配合使用效果较好。我国规定可用于饼干、肉禽罐头、调味料、糖果、蜜饯、凉果和饮料,按生产需要适量使用。
用途二:
本品具有特殊的甜味,其甜度约砂糖的250倍,从甘草中提取出来的甘草酸钠盐,即使稀释4000倍的水溶液也有甜味。但直接作为食品的甜味剂,对某些食品不合适,一般可与砂糖,葡萄糖、糖稀等天然糖类并用或与糖精、甘氨酸、丙二醇等适当配合,方可获得较为可口的甜味。日本1978年生产甘草酸240吨,原料甘草从我国和俄罗斯进口,最高年份,进口甘草12000吨,1978年销售总量中,用于医药、化妆品的占26%,用于食品的占70%,用于卷烟及其他的占4%,在食品方面主要用途是:1.酱油 除了可改善咸味以提高酱油固有味道外,可消除糖精的苦味,对化学调味剂有增效作用。2.咸菜类 与糖精并用腌制咸菜的卤法中,可消除糖精的苦味。在腌制过程中,可克服少加糖而出现的发酵败、变色、硬化等缺点。3.调味 本品可添加腌渍调味液,调味粉或饮食时的临时调味,可增加甜味,得到敌视芭的咸味,季低其他化学调味剂的怪味。4.豆酱 腌制小酱鲱鱼采用本品可起到增加甜味、使味道均匀的作用。在甘草提取的过程中,得到甘草酸铵溶解在含有讲学算量碳酸钠的水溶液中,减压浓缩即得甘草酸二钠(C42H60Na3O16)、甘草酸三钠(C42H59Na3O16)。也用作食品添加剂,作为豆酱、酱油的甜味剂。二钠盐对小鼠腹腔注射LD50为1.44克/公斤,三钠盐对小鼠皮下注射LD50为1.54克/公斤。
用途三:作为药物,具有抗炎、抗变态反应;作为甜味剂,广泛用于各类食品 

甘草酸药理药效

甘草酸具有抗炎、抗病毒和保肝解毒及增强免疫功能等作用。甘草酸具有肾上腺皮质激素样作用,能抑制毛细血管通透性,减轻过敏性休克的症状。可以降低高血压 病人的血清胆醇。由于甘草酸有糖皮质激素样药理作用而无严重不良反应,在临床中被广泛用于治疗各种急慢性肝炎、支气管炎和艾滋病。还具有抗癌防癌、干扰素 诱生剂及细胞免疫调节剂等功能。 

甘草酸生产方法

方法一:可由化学合成,但目前采用由甘草提取(见甘草次酸,前条)
方法二:将甘草的根茎干燥后粉碎至0.833mm的粉末(保留纤维部分),取粉末及纤维200kg,加水1200kg,在85~100℃下浸提2h。过滤后滤渣再用1000kg水提取2h,过滤后滤渣再重复浸提1次。合并3次滤液,在搪瓷蒸发器中浓缩至]/5的体积。冷却后加入95%的乙醇,使乙醇浓度达65%,静置24h,过滤除去植物蛋白、多糖等杂质。滤液中加入硫酸,调pH值至3,甘草酸沉淀析出。过滤、洗涤后,加入3倍的丙酮,加热回流3h,倾出丙酮提取液,残渣再反复回流提取2次。合并3次提取液,过滤后回收丙酮,即得湿甘草酸,于45℃干燥1h,缓缓升温至85~95~C,快烘干时,升至100~105℃,烘5min,经粉碎后即得成品。

甘草酸含量测定

鉴别:取本品0.2g,加水5ml,盐酸3ml蒸馏,于蒸馏液中滴加2,4-二硝基苯肼乙醇试液2~3滴,产生橙红色沉淀
检查:干燥失重 取本品1.0g,在80℃真空干燥8小时,减失重量不得过6.0%(中国药典2000年版二部附录VIII L)。
炽灼残渣:取本品1.0g依法检查(中国药典2000年版二部附录VIII N),遗留残渣不得超过0.2%。
重金属取炽灼残渣项下遗留的残渣,依法检查(中国药典2000年版二部附录VIII H第二法),含重金属不得过百万分之二十。
砷盐:取本品1.0g,依法检查(中国药典2000年版二部附录VIII J第一法),应符合规定(0.0002%)。
含量测定:取本品40mg,精密称定,置100ml量瓶中,加稀乙醇溶解并稀释至刻度,摇匀。精密量取稀释液10ml,置100ml量瓶中,加稀乙醇溶解并稀释至刻度,摇匀,照分光光度法(中国药典2000年版二部附录IV A),在252nm的波长处测定吸收度,按甘草酸(C42H62O16)的吸收系数(E1% 1cm)为141计算。

甘草酸测定方法

方法名称: 甘草浸膏粉—甘草酸的测定—毛细管区带电泳法
应用范围: 本方法采用毛细管区带电泳法测定甘草浸膏粉里甘草酸的含量。
本方法适用于中药制剂甘草浸膏粉。
方法原理: 用毛细管区带电泳法,紫外检测波长228nm ,电压14kV ,以50mmol/L硼砂溶液为背景电解质,氢噻嗪为内标,对甘草浸膏粉中的甘草酸进行定量分析。

甘草提取甘草酸的工艺流程 

1.粉碎:取甘草根、茎在粉碎机中粉碎,过20目筛备用。

2.提取:取上述甘草粗粉200kg,放入2000L的搪瓷反应釜中,加水1200kg,缓缓加温到85~100℃,搅拌提取2h,过滤,滤渣 再加水1000kg,按上法提取2次,合并3次滤液。

3.浓缩:将提取液用泵抽入搪瓷蒸发器中进行浓缩,当浓缩 到原体积的1/5时,趁热过滤。

4.分离:滤液冷至室温加95%乙醇使提取液中的乙醇含量 达65%,静置24h,然后过滤,除去植物蛋白、多糖。

5.沉淀:取甘草滤液,放入沉淀池,加入浓硫酸调pH=3,甘 草酸则沉淀析出,过滤,用去离子水洗2次,然后真空抽干。

6.精制:在甘草酸沉淀中加3倍量的丙酮,加热回流提取 3h,过滤、残渣再反复回流草取2次,合并3次回流提取液,过滤, 回收丙酮,得甘草酸湿品。

7.干燥:将干草酸湿品放入烘房干燥,开始控制温度在 45℃,干燥1h,缓慢升温到85~95℃,近烘干时,升温到100℃,最 后在105℃烘烤5min。

8.粉碎:将烘干的甘草酸放入粉碎机中粉碎,过60~80目筛,即得甘草酸粉末。


甘草酸衍生物

    在甘草酸的糖配体上引入甘氨酸乙酯,其凝血素和溶血素分别提高了近3倍和2倍,而保持30位羧基裸露,在糖苷上引入缬氨酸叔丁基后,其溶血素类水平比30位羧基保护提高了50%,甘草酸的糖配体上引入氨基酸和短肽后其抗SARS病毒的效果比甘草酸平均提高10倍左右,特别是在糖苷键引入甘氨酸和赖氨酸短肽,其抗 SARS 病毒的效果比甘草酸提高70倍。

精密称取甘草酸单铵盐4. 2mg(相当于甘草酸13. 9mg) ,置25ml量瓶中,用50mmol/L硼砂溶液定容至刻度,超声20min作对照品溶液。
2.氢氯噻嗪溶液的制备
精密称取氢氯噻嗪0.2537g ,置250ml 量瓶中,用甲醇定容至刻度,摇匀,即得。
3. 标准溶液的制备
精密取对照品溶液适量,置10ml量瓶中,加入氢氯噻嗪溶液1. 0ml ,用50mmol/L硼砂溶液(含体积分数10 %甲醇) 定容至刻度,摇匀,配成浓度为0. 1390 、0. 2780、0. 5560、1. 1190、2.2380 g/L,作为绘制标准曲线的系列浓度标准溶液。
4.供试品溶液的制备
精密称取甘草浸膏粉约0. 113g ,置50ml 量瓶中, 另精密加入氢氯噻嗪溶液5. 0ml , 用50mmol/L硼砂溶液(含体积分数10 %甲醇) 定容至刻度,,超声30min,滤过即得。
注:"精密称取"系指称取重量应准确至所取重量的千分之一。"精密量取"系指量取体积的准确度应符合国家标准中对该体积移液管的精度要求。

操作步骤
1.标准曲线的绘制
取上述标准溶液在毛细管电泳仪上进样10s,记录电泳图10μL注入高效液相色谱仪,用紫外检测器测定甘草酸的吸收度,以标准溶液浓度对峰面积绘制标准曲线,进行线性回归,得到回归方程。
2. 供试品的测定
取上述供试品溶液在毛细管电泳仪上进样10s,用紫外检测器测定甘草酸的吸收度,计算出其含量。

甘草酸在各个行业的应用比例如下:用于医药化妆品占26%,用于食品的占70%,用于卷烟及其他的占4%。
